God was preparing Moses for leadership and for spiritual warfare with Pharaoh to subdue the powers of darkness and free God's people. The rod was a symbol of authority and spiritual power used to perform miracles, signs and wonders.
When God anointed Moses with the Holy Spirit boldness entered his spirit and he was empowered to take on his mission to deliver God's people! When I was baptized in the Holy Spirit boldness permeated me, fear, doubt and unbelief left and a new power came upon me to preach, teach and eventually write.
Moses was about to see the signs of God's judgment poured out on Egypt, so He demonstrated His power by making Moses hand leprous and then healing it as well as turning his rod into a snake. When God's anointing is given one must step into it and walk in that which has empowered him.
And you shall take this rod in your hand with which you shall do these signs. Exodus 4:17
